What is the Red Blood Cell Lysing Reagents?

Red blood cell lysing reagents selectively lyse erythrocytes in whole blood or bone marrow while preserving leukocyte scatter and antigen expression. They are used in no-wash or wash protocols for immunophenotyping, and come as ammonium chloride-based lysis buffers or fixative-containing lyse-fix formulations.

Overview

Whole blood contains about 1,000 red blood cells per white blood cell, which would overwhelm a flow cytometer if not removed. Lysing reagents exploit differences in erythrocyte membranes: ammonium chloride based solutions, used at about 0.15 M with bicarbonate and EDTA, take advantage of the carbonic anhydrase pathway to swell and lyse erythrocytes within 10 minutes while leaving leukocytes largely intact. Lyse-fix reagents combine a mild fixative, typically formaldehyde, with a lytic agent for a single-step, no-wash procedure that stabilizes samples for up to 24 hours. Protocol choice affects lymphocyte scatter separation, leukocyte viability and loss of fragile cells such as plasma cells or blasts. Ready-to-use or 10x concentrates are available, and lysing time and temperature are critical, since over-lysis causes debris and leukocyte loss.

How does it work?

Cytometry reagents make cellular properties detectable as optical signals. Monoclonal antibodies conjugated to fluorochromes such as FITC, PE, APC, or tandem and polymer dyes bind specific antigens, and the laser-excited fluorescence is proportional to antigen density. Nucleic acid and amine-reactive dyes discriminate live from dead cells, because membrane-compromised cells take up DNA dyes or expose more amines. Calibration and compensation beads bind antibodies through capture moieties or carry defined fluorochrome content, providing consistent positive and negative populations for compensation matrices and standardization. Fixation cross-links proteins, often with formaldehyde, while permeabilization with detergents or alcohol allows antibodies to reach intracellular or nuclear targets. Lysing solutions remove red blood cells by osmotic or ammonium-chloride lysis while preserving leukocyte scatter properties.

What are the key features?

  • Selective erythrocyte lysis preserves leukocyte scatter and surface antigen expression
  • Ammonium chloride formulation for wash-based protocols and viable cell recovery
  • Lyse-fix formulation enables a single-step no-wash workflow
  • Samples stabilized for up to 24 hours after lyse-fix treatment
  • Available ready-to-use or as 10x concentrate in 100 mL to 1 L bottles
  • Suitable for whole blood, bone marrow and cord blood specimens

What are the technical specifications?

A typical red blood cell lysing reagents configuration offers lysis chemistry of Ammonium chloride / lyse-fix (formaldehyde), working concentration of 1× (10× stock available), lysis time of 10–15 min at room temperature, sample-to-lysing reagent ratio of 1:10 to 1:20 (v/v), wash step of Required (ammonium chloride) / optional (lyse-fix) and post-lysis stability of Up to 24 h (lyse-fix). Values are indicative of the product class; the exact figures for each configuration are confirmed on the quotation and the calibration documents supplied with the instrument.

Which applications is it used for?

The Red Blood Cell Lysing Reagents is used in lymphocyte subset whole blood assays, leukemia and lymphoma immunophenotyping, CD4 and CD34 counting, bone marrow sample preparation and high-throughput no-wash staining. Across the wider Cytometry Reagents class, laboratories also deploy it for immunophenotyping panels, live/dead cell discrimination and instrument compensation and QC.

Which standards and certifications apply?

Relevant standards and methods include CLSI H62, CLSI H43, ISO 20391-2, ISO 13485 (IVD reagent manufacturing), EU IVDR 2017/746 and ISAC MIFlowCyt. Certification candidates for this model are ISO 13485 manufacturing, CE-IVD marking for selected products and ISO 9001 manufacturing; each is to be confirmed for the specific configuration before shipment, and certificates are supplied only where they have been issued.

Store antibodies and dyes at 2 to 8 degrees Celsius, protected from light, and never freeze tandem dyes, which degrade and cause spillover. Titrate every new antibody lot, and record lot numbers in your experiment records. Use Fc receptor blocking where needed, and include fluorescence-minus-one and isotype or biological controls when setting gates. Prepare fresh formaldehyde solutions and validate fixation time for each epitope. Dispose of azide-containing buffers and lysing reagents according to local chemical regulations, as sodium azide can form explosive metal azides in plumbing. For regulated assays, source reagents with certificates of analysis, and revalidate panels when changing vendors or clones.

How do I select the right Red Blood Cell Lysing Reagents?

Choose ammonium chloride lysis when viable leukocytes or later functional tests are needed, and lyse-fix reagents for fast, stable no-wash clinical workflows. Fragile populations may need gentler lysing. In an RFQ, specify the concentration, specimen type, protocol (wash or no-wash), bottle size and whether IVD status is required.

How does it compare with other Cytometry Reagents variants?

Choose reagents to match the instrument's lasers and filters, since fluorochromes must be excited and detected by your configuration. Assign bright fluorochromes to low-density antigens and dim ones to abundant antigens, and avoid spectral overlap between tandem dyes. Confirm antibody clone, species reactivity and validated application for flow cytometry. For intracellular or phospho targets, select compatible fixation and permeabilization buffers. Include a viability dye in every panel, and use compatible compensation beads for each fluorochrome. Check lot-to-lot consistency documentation, storage conditions at 2 to 8 degrees Celsius, expiry dating and supply of bulk or lyophilized formats for regulated assays. Titrate each antibody in your own cells to find the optimal staining concentration.

Frequently asked questions about Red Blood Cell Lysing Reagents

A lysing reagent such as ammonium chloride removes red blood cells but does not fix leukocytes, so samples are processed promptly. A lyse-fix reagent adds a fixative to stabilize leukocytes in one step, allowing delayed acquisition for up to about 24 hours.
